Overview

At Massey University, we are currently looking for a DNA Synthesiser which meets below minimum requirements:

The DNA synthesiser should be able to synthesize from 4 to 12 standard or modified oligos in a single run, using standard or modified phosphoramidite chemistries. The total synthesis run time for 12 simple PCR quality 20-mer oligos should be less than 3 hours. In the standard operating mode, the DNA synthesiser should be capable of making 50 nmol scale oligos up to 80 bases in length with coupling efficiencies upwards of 99%. The DNA synthesiser should be capable to handle longer oligos (>120mers), and different scales (50 nmol and up to 200 μmol) within a single run, which should allow great user flexibility. Columns may be added and removed during a run. The DNA synthesiser should be equipped with Trityl monitors to monitor coupling efficiency during DNA synthesis. The instrument should allow automatic delivery of phosphoramidites for wobble nucleobases with no premixing of amidites. There should be at least 8 amidite positions and at least 6 reagent positions. Software should allow changes in the synthetic protocols.
